Introduction
All the primer design is made based on the assumption that Gibson ligation or any other method using overhands is used.

Once you have chosen one or several DNA fragments to insert into a vector you have to design the primers that will allow you to ligate or in other words to connect your fragment to your vector in order to generate your plasmid.
